SUMMARY:Adipose Biology Seminar Series
DESCRIPTION:The adipose tissue is an organ widely distributed throughout the body\, and it is involved in a large variety of medical conditions and metabolic disorders. In recent years\, there has been a growing interest in adipose tissue biology research\, both internationally and locally\, in different departments and institutes. This seminar series serves to facilitate discussion and foster collaborations between PIs and trainees with an interest in adipose tissue research in the Toronto region and beyond. The Adipose Biology Seminar Series is an online monthly meeting in which PIs and trainees will present and discuss novel data pertinent to adipose tissue biology. We aim to host speakers from diverse research foci such as obesity\, diabetes\, cancer and trauma to provide an opportunity to learn about exciting new developments in the field and build connections within a local network. To sign up and receive information about upcoming meetings\, please contact Laura Prochazka. \n\nLearn more about Medicine by Design discussion groups\n\nTalk Abstract\nCRISPR-Cas technology has revolutionized genome editing and has facilitated the elucidation of gene function and therapeutic targets for diseases such as cancer. During my PostDoc in Jason Moffat’s lab I have developed and applied genome-wide CRISPR-Cas9 screens for the identification of fitness genes in human cells. More recently\, I have applied CRISPR screens for the global mapping of genetic interactions for genes functioning in cellular metabolism\, whose rewiring is a cancer hallmark and a major focus in the development of novel therapeutic agents. Our data reveals extensive cross talk between metabolic pathways and uncovered the functional role of previously uncharacterized genes. Finally\, we have developed a novel CRISPR-Cas screening platform that allows for highly specific combinatorial targeting of multiple genes\, genetic elements or genomic sites. We have applied this platform to reveal extensive genetic interactions between paralogous genes and uncovered previously masked phenotypes due to genetic redundancy. \nCollectively\, in this seminar I will present the CRISPR-based screening strategies we have generated to map genetic interaction networks in human cells that can be leveraged to study the wiring of cellular states\, determine novel gene functions and serve as a source for the rational design of combinatorial anti-cancer therapeutic strategies.

To sign up and receive information about upcoming meetings\, please contact Laura Prochazka. \n\nLearn more about Medicine by Design discussion groups\n\nTalk abstract\nThe implementation of colorimetric detection techniques has offered a powerful means of producing rapid qualitative results with easy visual readout. However\, these techniques are usually limited to either a “yes” or “no” answer and have difficulty multiplexing over three targets simultaneously. Electrochemical detection is capable of providing solutions to these limitations. The key advantage of electrochemical assays is the ability for signal amplification\, allowing for trace analyte detection. Recent literature has highlighted the promising avenues of synthetic biology and cell-free gene circuits. This project is the first of its kind to detect cell-free synthetic gene circuits through electrochemical readout. A gold-based electrochemical sensor has been designed to implement five different detection sites for restriction enzyme cutting of surface-immobilized DNA labelled with methylene blue. Restriction enzymes are produced through a cell-free toehold gene circuit and are easily redesigned for disease-specific detection. By coupling electrochemical detection and cell-free toehold gene circuits\, we were able to create a multiplexed assay for applications in antibiotics and antibiotic resistance detection.

To sign up and receive information about upcoming meetings\, please contact Laura Prochazka. \n\nLearn more about Medicine by Design discussion groups\n\nTalk Abstract\nThe field of DNA nanotechnology uses nucleic acids as a “brick & mortar” toolset to build synthetic molecular structures and devices. These DNA-based tools leverage the sequence-programmability and predictability of nucleic acid hybridization to enable precise spatiotemporal manipulation of macromolecules as well as higher-ordered assemblies. A central motivation of the field is to be able to design and create artificial molecular assemblies that rival the complexity of natural molecular systems\, and that can perform user-defined tasks across molecular to cellular length scales. The pursuit of this goal also aims to contribute towards a bottom-up understanding of how the spatiotemporal dynamics of biomolecular assemblies dictate their function\, which can inform synthetic biology design. \nIn this talk I will provide an overview of DNA nanotechnology and briefly introduce its fundamental as well as emerging capabilities. I will highlight existing examples of DNA-based molecular tools and devices\, including their use for synthetic biology applications\, such as for controlling receptor signaling and gene expression. I will also highlight efforts from our own group to engineer bio-inspired molecular devices for applications in biological sensing and therapeutic discovery.

Modelling these circuits is essential to both explaining how they work and informing the forward design of novel systems with desired dynamic responses to signal inputs. Beyond this\, new dynamic responses may be possible with the rational\, model-driven repurposing of biological parts. In this tutorial\, the basic steps of dynamic modelling will be presented along with several resources to assist you in further exploring model-based design in your synthetic system. All skill levels are welcome and encouraged to attend.

DESCRIPTION:Trevor McKee\, PhD\, will speak on “Moving from Images to Data: Resources and expertise in computational pathology at the STTARR core facility.” \nThis group is open to investigators\, trainees and staff affiliated with the University of Toronto. To sign up to get more information on upcoming discussion groups\, please contact Laura Prochazka. \n\nLearn more about Medicine by Design discussion groups\n\nTalk abstract \nA major bottleneck in synthetic biology approaches to medicine is the lack of quantitative data on tissue morphology to inform structural information on pathological and healthy tissue states. Excitingly\, the advent of digital pathology with increasing semi-automated computational analysis workflows is moving the discipline towards a quantitative science. Computational pathology is providing new tools that can both augment a clinical pathologist’s toolbox\, as well as provide new insights into tissue structure at single-cell resolution to guide the development of synthetic biology-guided next-generation therapeutics. Single-cell resolution structural and positional information is also a major part of the Human Cell Atlas\, a worldwide consortium to map every cell and cell lineage in the developing and adult human. We are therefore delighted to gain some insight into the work of Dr. Trevor McKee\, visiting from STTARR to talk to us about “Moving from Images to Data: Resources and expertise in computational pathology at the STTARR core facility.” \nDr. McKee is a biological engineer who received his PhD from MIT under the guidance of Dr. Rakesh K. Jain\, on the application of intravital microscopy to studying barriers to drug and gene delivery in tumours. He runs the Computational Pathology core facility within STTARR\, a multi-modality imaging facility within the Princess Margaret Cancer Centre\, where he leads a team of programmers and technicians to build algorithms to quantitatively measure protein and RNA abundance at the single-cell resolution in whole-slide pathology images\, for academic and pharmaceutical clients. \n
